In Myanmar, potato is also one of the main culinary crops and a promising profitable crop to growers, so cultivation of potato has been being extended to the arable lands. Mass production of disease free, high quality seed tubers for potato growers is demanded. Moreover, rapid and economically feasible tissue culture technology for potato shoot multiplication needs to be explored. The experiment was conducted at the Plant Tissue Culture Laboratory of Department of Horticulture and Agricultural Biotechnology, YAU from 2015 to 2016. The experimental design for this experiment was 3 factors factorial in RCB arrangement with 4 replications. Factor A was three potato cultivars, Up-to-date, L-11 and Atlantic, and factor B was three different combinations of plant growth regulators(PGRs), 0.05 mg.L-1 Naphthaleneacetic acid (NAA) combined with two different levels of 0.5, 1.0 mg.L-1 6-benzyl aminopurine (BAP) and PGRs free. Two different culture systems, liquid culture and solid culture system were used as factor C. Liquid culture system was superior to solid MS in such growth parameter as shoot length, no. of nodes, no. of roots and fresh weight. Free PGRs provided the best result in liquid MS medium.
