Nowadays, tissue culture technique has been widely\r used for most rare species and species that are difficult to propagate Therefore, tissue\r culture became an important method for propagation of Phalaenopsis. Tissue culture\r techniques, explant types, plant growth regulators and culture systems are mainly\r attributed to success. Two experiments were carried out by using different flower\r stalk nodes at different positions and in vitro young leaves. The objective of the\r research is to investigate the effect of plant growth regulator, culture systems and\r media in in vitro regeneration.\r In flower stalk node culture, the effects of 1st, 2nd, 3rd, 4th, 5th and 6th nodal\r positions counted from base to top were tested for shoot regeneration. In leaf culture,\r two culture systems (solid and cotton support medium) and different concentrations\r and combinations of benzyladenine (BA) 2 and 4 mg.L-1, thidiazuron (TDZ) 1 and\r 2 mg.L-1, with and without naphthaleneacetic acid (NAA) 0.5 mg.L-1 were tested for\r protocorm like bodies (PLBs) induction from leaf explant. In PLB proliferation stage,\r three culture systems (solid, liquid and cotton support liquid culture medium) were\r tested. Vecin and Went (VW) and Murashige \u0026 Skoog (MS) basal medium with and\r without 0.1 mg.L-1 BA and 1 mg.L-1 NAA were tested in rooting stage of shoot\r developed from leaf culture.\r Among the flower stalk node positions, better responses were observed from\r 2nd nodal position in shoot multiplication. 3rd nodal position gave best results in root\r regeneration and hardening stages. Using flower stalk node culture for mass\r propagation, 2nd, 3rd and 4th middle nodal positions should be used as the explants for\r production of Phalaenopsis amabilis orchid. In leaf culture, maximum number of\r PLBs per explant (7.1) was observed from medium supplemented with 2 mg.L-1 BA.\r Culture systems used in initial culture were not statistically difference among each\r other. But solid culture showed better results than the cotton culture. The interaction\r effect between PGR and culture systems was observed in survival % and number of\r PLBs per explants. The results indicate the used of PGRs vary according to the\r culture systems. Therefore, BA should be used in cotton culture and TDZ in solid\r culture. Cotton culture gave the best result in PLBs proliferation on leaf explants.
